The city beat off competition from Dublin and Bilbao in order to take the accolade.
Roshene Singh, chief marketing officer of South African Tourism, said it is "wonderful news" and shows that the nation has some of the best creative minds in the world at the moment.
Events held over the past year such as the Cape Town International Jazz Festival, the Joy of Jazz and Macufe Festivals and the Joburg Art Fair served to highlight the qualities on show.
The decision to award the title to Cape Town was taken by the International Council of Societies of Industrial Design, an organisation founded in 1957.
Torino in Italy, Seoul in South Korea and Helsinki in Finland have all been given the accolade in the past.
